چکیده مقاله:

Methamphetamine is a neurotoxin and potent psycho-stimulant of the phenethylamine and amphetamine classes that is used to treat attention deficit hyperactivity disorder (ADHD) and obesity. Methamphetamine exists as two enantiomers, dextrorotary and levorotary. Dextromethamphetamine is a stronger central nervous system(CNS)stimulant than levomethamphetamine; however, both are addictive and produce the same toxicity symptoms at high doses. We have studied Metamphetamine affects on gene expression in individuals who are addicted to this drug and used it forseveral time in more than one year. blood samples collected from 30 addicted and 30 normal person in the same range ofage. RNA of whole blood extracted by column method kit . cDNA synthesized and expression of 300 gene investigated with microarray (Affymetrix gene chips) technique. expression of 12 gene had been affected . over expression of NDUFS1 and NDUFS2 , and decreasing of expression in 5HT2a,DRD2,DRD3,BDNF,DISC,NRG1,MAOA,bcl2,NDUFV1 andNDUFV2. Microarray results for two gene NDUFS1 and NDUFS2 confirmed by Real Time PCR syber green method andresults analyzed by SPSS and Bonferroni statistical soft wares. In our samples a significant affects of Metamphetamin on expression of several genes especially in mitochondria complex 1 subunits genes have been detected
