A Comparative Analysis to Evaluate the Drug Response of Breast Cancer Cell Cultured in 2D and Scaffold-Based 3D Cell Culture System

Breast cancer is a tumor arising from breast cells. Resistance has been described in terms of the available treatments; surgery and radio/chemotherapy which may result from microenvironment or pharmacological factors. 3-dimensional (3D) cell culture is an artificially created environment in which cells are allowed to grow and interact with their surroundings. Therefore, studies using this method help to look more closely at the cell behavior rather than 2-dimensional (2D) culture. Herein, we studied the growth rate of two breast cancer cell lines, MCF-7, MDA-MB231, on the matrigel as a well-known scaffold for 3D culture. we examined their drug responses in comparison to 2D. The cells were cultivated in 3D (On-top and Embed manners), the mammospheres were generated then the growth curve was studied. Afterward, in the separate experiments the cells were treated with two well-known pro-apoptotic drugs, paclitaxel and actinomycin-D, at the IC50 value determined by MTT test. Subsequently, the cell-growth was re-evaluated. Results show that growth in 2D and 3D-Embed methods have the similar rates but it is higher for momammosheres in 3D-On-top. Besides, size of the generated mammospheres from MCF-7 is larger than MDA-MB231 in 3D cultures. Furthermore, in 2D culture the drug responses of two cell lines are same whereas in 3D culture, they vary according to the cell type, culture method and the anticancer drugs. Regarding to the observed variety in parameters affecting the cellular behavior, it is concluded that 3D is an appropriate replace for 2D cell culture in drug response studies.
