Computational Investigation of Hsa-mir-196a-2 Targetome Pathway and its SNP Function in Patientswith Gastric Carcinoma: Probable Association Between Alleles and Patients Prognosis

Gastric cancer (GC) is the third most common cancer and the second leading cause ofcancer fatality worldwide. In 2012, about three quarters of novel GCs happened in Asia.Helicobacter pylori infection is an important risk factor for GC. MicroRNAs (miRNAs) aresmall noncoding RNAs that work as post-transcriptional regularizers of gene expression andinvolve in various cancers. Single nucleotide polymorphisms (SNPs) which are located inmiRNAs locus may affect either expression or function of miRNAs. The miR-196a-2polymorphism rs11614913 is related to the susceptibility of diverse tumors, including gastriccancer.This study aimed to expand current knowledge about molecular function of miR-196a-2 and its SNP in GC cells as a prognostic biomarker in GC patients. MiRNASNP databaseillustrates SNP in miR-196a-2. MiRBase database gives general information about miR- 196a-2. Predicted targets of miR-196a-2 were obtained from miRWalk database. Targetomeexpression in GC was evaluated in UniGene database. Finally, DAVID database was usedfor molecular pathways enrichment analysis. It is predicted that miR-196a-2 functions as acrucial tumor suppressor by inhibiting some significant genes in epithelial cell signaling inHelicobacter pylori infection such as PAK1, JNK, IκBα that these genes play a vital role inchemotactic effect. Chemotactic activity quickly increases Helicobacter pylori colonizationand infection. Furthermore, T allele causes the stability reduction of miR-196a-2 structure.Subsequently, there is a probable connection between T allele and poor prognosis of GC. Inother words, rs11614913 may contribute to increase Helicobacter pylori infection and GCsusceptibility due to alteration of epithelial cell signaling. According to our data, miR-196a-2and its SNP may be involved in GC and its prognosis by altering regulation of epithelial cellsignaling in Helicobacter pylori infection and some key signaling pathways. Shortly, T allelein this position can have prognostic value for GC.
