چکیده مقاله:

The cotton bollworm, Helicoverpa armigera (Hübner) is one of the serious pests of differenteconomic crops in many parts of the world including Iran. Study of the insect digestive enzymes isnecessary in order to reach practical and new pest management technologies. The midgut from H.armigera fifth instar larvae reared on artificial diets based on various chickpea varieties (Arman , Azad ,Binivich and Hashem) and one cowpea variety (Mashhad) were anatomized, and supernatants were usedas the source of enzyme. Results of zymogram analysis on gel showed that protease enzymes in midgut ofthe cotton bollworm fed on different chick pea and cowpea varieties had two isoforms. The km values of0.661, 0.048, 0.246 and 0.054 mM and the Vmax values of 0.578, 0.204, 0.293 and 0.174 mmol/min wereestimated for trypsin on Arman, Azad, Binivich and Hashem, respectively. Also calculated Km and Vmaxfor chymotryipsin were 0.032 mM and 0.442 mmol/min, respectively. Determining biochemicalcharacteristics of insect digestive enzymes could be useful to reach safe and suitable controlling methods,as well as, to obtain better understanding of the enzymes role in nutritional physiology of the insect pests
