Targeting of mannosylated meglumine antimonate-loaded albumin nanoparticles to Leishmania major-infected macrophages for the treatment of cutaneous leishmaniasis

Introduction: First-line drugs for treatment of Cutaneous Leishmaniasis (CL) is pentavalent antimony compounds such as Meglumine Antimonsate (MA), although they are toxic and have several side effects. Drug delivery system can be used to send the drug to Leishmania-infected macrophages and in such conditions, many of the problems associated with the conventional drugs can be solved. In this study, we evaluated anti-leishmanial activity of meglumine antimonate-loaded albumin (MA-Alb-NPs) and containing mannose nanoformulations (MA-Alb.Mann-NPs) on Leishmania major-infected macrophages compared with the free drug.Material & Methods: MA-Alb and MA-Alb.Mann nanoparticles were prepared by Precipitation and Millard reaction methods using albumin as carrier and Mannose as surface ligands. After evaluating designed nanoparticles, cytotoxicity study was performed to determine the CC50 of nanodrugs on J774 macrophages. Anti-leishmanial effects were evaluated on experimentally Leishmania major-infected macrophages by calculating IC50. All tests were performed as triplicate.Results: The IC50 values of MA-Alb-NPs, MA-Alb.Mann-NPs and MA were calculated 22.78 ± 1.35, 12.53 ± 1.09 and 109.1 ± 2.03, respectively. The parasitic load in the control group was obtained 2.60, while this value was 2.16 at the lowest concentration of MA-Alb.Mann-NPs (0.12μg/ml) and by increasing the drug concentration, the parasitic load has also decreased, as at the highest concentration (90μg/ml), it has fallen to 0.13.Conclusion: The results showed that both nanodrugs had a significant antileishmanial effect on the Leishmania major-infected macrophages compared with the control drug. However, the greatest reduction in parasitic load is related to MA-Alb.Mann-NPs. Therefore, this form of drug can be a good alternative to the current medications and it is suggested that further studies should be done in vivo in this regard.
