Objective: The main aim of the present study was to investigate the ASCs’ effect on radiation induced skin damage in rat.Method and Materials: In this experimental case-control study, we examined 4 groups of rats each containing 8 female rats (weighing 180-250 gr and 8 weeks of age). The rats received a 30 Gy single dose to their buttocks skin at a dose rate of about 0.28 Gy/min.Twenty-four hours after irradiation, stem cells were transplanted subcutaneously and intravenously in each half of the case group and control group has no transplantation. The rats were examined and observed by microscopic analysis, phenotypic score (0, 1, 2, 3) and histological assay during 8 weeks of follow up, weekly.Results: The wound size in the control group was significantly higher than case group in the second, third and fourth weeks of evaluation (P < 0.05). There was not any significant difference between two groups in skin lesion severity (P > 0.05). There was not any significant difference between IV and local groups in wound size or skin lesion severity (P > 0.05). There was not any significant difference in pathological factors and onset of recovery signs between two groups (P > 0.05).Conclusion: Accordingly, ASCs’ ability for differentiation to epithelial cells is probably lower compared to the other sources of MSCs, so further investigations comparing different sources are recommended.
