Evaluation of the Effect of Using Pericardial Membrane and Xenograft Umbilical Mesenchymal Stem Cells on Bone Healing in Rats

Intruduction: Incomplete or complete detachment of the bone structure is called bone fracture, which is associated with some degrees of soft tissue irritations. Diagnosis is based on history, clinical symptoms, and X-rays. Bone fractures have many different types; each with different symptoms and images. Bone healing is unique because it is not the result of scarring, but the injured tissue is actually repaired. Bone repair has many steps. One way to speed up this process is to reduce the time period of each of these steps. There are many ways to accelerate this process and the use of pericardial membrane and umbilical cord mesenchymal stem cells is one of the newest methods.Material and Method: This experimental study was performed on 30 adult male Wistar rats weighing approximately 250-300 grs. All stages of this study were designed and implemented in accordance with the ethics protocols of the International Laboratory Committee for the Protection of Laboratory Animals.Rats were randomly divided into three groups:Group I: Bone defect was created and left empty. (Control group)Group II: After bone defect was created, the defect was filled with pericardial membrane. (Pericardial membrane group)Group III: Bone defect created was filled by pericardial membrane and umbilical cord mesenchymal stem cells. (Pericardial Membrane and Stem Cell Group)After 45 days, the mice were euthanized. Biopsy and histopathological examination was performed.Results: By evaluating the histomorphometry results, we found out that the amount of lamellar bone formed in the group treated with pericardial membrane was significantly higher than the control group and the group treated with pericardial membrane and stem cells (P<0.05). No significant differences were observed in biomechanical experiments.Conclusion: This study showed that the use of pericardial membrane can be effective in increasing bone mineralization in the defect site.
