چکیده مقاله:

Background:Mesenchymal stem cells (MSCs) are found in bone marrow and adipose tissues. It have beenindicated that these cells migrate to tumor areas and secrete angiogenic factors such asVEGF-A that contribute to tumor angiogenesis. Based on the known effects of arachidonicacid (AA) metabolites on over expression of VEGF-A in other cell types and in order toclarify role of AA in tumor angiogenesis through its effect on MSCs we investigate the effectof AA on CXC chemokines, that their impact on the process of angiogenesis has beendemonstrated, as well as VEGF-A and IL-6 were evaluated in MSCs.Materials and Methods:After isolating MSCs from the adipose tissue by collagenase in order to identify genesaffected by AA from Real-Time RT-PCR method to investigate the expression of CXCL-1 toCXCL-17 (except CXCL-15), and from ELISA method to investigate secreted VEGF-A, IL-6and CXCL-8 were used. Also, the MTT assay and Scratching assay was used to evaluate theeffects of AA on cell proliferation.Results:Real-Time RT-PCR showed increased expression of CXCL-8, CXCL-12 and decreasedexpression of CXCL-4. ELISA results also indicate an increase in the secretion of VEGF-A,IL-6 and CXCL-8. Although the MTT assay showed no significant change in cell survival butScratching assay showed increased cell proliferation at high concentrations of the AA.Conclusion:It can be inferred that AA can stimulate tumor angiogenesis by MSCs via stimulating theangiogenic factors production of tumor area migrated MSCs that is very important inmalignant tumor creation.
