Background and Objective: In recent years, the synthesis of neutral lipids linked to crown ethers able to coordinate cations and form stable complexes with DNA could represent an important strategy in gene therapy. In 2013, Angelini et al. introduced neutral liposomes containing crown ether-lipids as potential DNA vectors. Their main goal was to evaluate two synthesized crown ether lipids 1,2-O-dioleoyl-3-O-{2-[(12-crown-4)methoxy]ethyl}-sn-glycerol (12-crown-4L) and 1,2-O-dioleoyl-3-O-{2-[(15-crown-5)methoxy]ethyl}-sn-glycerol (15-crown-5L) as new liposomes that have specific drug delivery capabilities. Material and Method: All materials needed for the synthesis of crown ethers and liposomes were purchased from Avanti Polar Lipids and Sigma-Aldrich. The method of synthesizing crown ethers is presented in Scheme . Scheme 1. a) NaH 60%, dry THF; b) LiAlH4, dry THF, RT; c) MsCl, Et3N, DCM; d) NaH 60%, dry THF; e) p-TsOH, MeOH; f) Oleic acid, DCC, DMAP, DCM, overnight at roomtemperature Findings: Dynamic Laser Light Scattering measurements after the extrusion shown that the incorporation of the guests tends to decrease the mean diameter of POPC liposomes, second to the concentration of12C4L, and 15C5L into the bilayer (Table 1). Conclusion: The results of their research showed that the crown ethers in the guest molecules promoted the interaction of the bilayer with DNA, respectively, and the capacity of the investigated liposomes in interaction with DNA in presence of Ca2+ was confirmed by SAXS experiment. Also, mixed liposomes were able to complex DNA in the presence of Ca2+ and could provide a basis for designing and employing new systems as DNA carriers.
