The effect of hydroalcoholic lavender (Lavandula officianalis) extract on NT3 gene expression after sciatic nerve compression in rats

Retrograde transport to the cell bodies of alpha motoneurons after the nerve lesion causes spinal degeneration. The neurotrophic factor (NT3) increases the number of myelinated axons in the dorsal root during the recovery process which is necessary for corticothalamic connections. Also it leads to both differentiation and survival of sensory neurons, parasympathetic neurons and motoneurons and prevents cell death. Lavender is a plant in the family Lamiaceae. While some researches have reported antioxidant, antispasmodic, diuretic, anti-asthmatic, and antipyretic effects of the plant, therefore, this study examined NT3 gene expression changes on 28 day after sciatic nerve compression in rats, in the presence of Lavandula officinalis extract. Lavender Soxhlet hydroalco-holic extraction was prepared. In this study, 36 male Wistar rats (200-250 g, 3 months old) were provided, then randomly divided into 3 groups include control, compression and treatment (Compression group + hydroalco-holic extract of Lavender injections 75mg/kg) groups. In controls the muscles was opened without damage to gain access to the sciatic nerve without damaging. In compression and treatment group the sciatic nerve (right leg) was compressed for 60 seconds. Two occasions of the extract was injected intraperitoneally. A biopsy was taken from the spinal cord segments L4-L6 on day 28. after that total RNA was extracted, cDNA was synthesized and NT3 gene expression changes were studied in control, compression and treatment groups. The results showed that NT3 gene expression had a significant reduction in compression group compared to the control group. The NT3 gene expression had a significant increase in treated group (Compression + hydroalcoholic extract 75mg/kg) compared with the compression group. A significant increase in gene expression shows that Lavandula officinalis hydroalcoholic extract that has Antioxidant and anti-inflammatory properties can improve nerve regeneration via NT3 gene expression.
