چکیده مقاله:

In order to gene cloning, at first desired gene should separate the from other contaminants (proteins, primer-dimer bands unskilled, etc.) and then the purified gene was used for recombinant plasmid from E.coli bacteria. To date, various physical and chemical methods is used for purification of target gene of other gene contamination, But in this paper, a new physical methods using for gene purify to be used by the electric field within TAE.1X fluid. Given that in order to interest plasmids recombination and gene cloning, purified gene needs to be Ligate to bacterial plasmid that this ligation depends on the gene solvent environment. until now, chemicals conventional methods used of double distilled water to dissolve the gene was purified by agarose gel But in this method to solve the genes were purified from agarose gel, from TAE.1X is used instead of double distilled water, Finally, the ability of ligation purified gene to plasmid and also ability of cloning this gene was tested by E.coli strain DH5α. In this study, PqHMGR gene (FJ755158.1) amplification was used by PCR using primers designed from the cDNA sequences of HMG-COA reductase of the ginseng plant (Panax quinquefolius).Finally, PqHMGR gene was purified device and procedure and well was cloned by DH5α strain of E.coli bacteria correctly. In order by the foresaid .to confirm the successful cloning and sequencing, cloned recombinant plasmids was sent to Bioneer Company
