The effect of vitamin C on structure and function of ovaries following embedding in platelet-rich-fibrin bioscaffold and autotransplantation in the gluteussuperficialis muscle

Chemo and radiotherapy in cancer treatment often lead to ovarian failure and infertility. Ovary transplantation which is an effective way to preserve fertility in these patients is accompanied by ischemic-reperfusion injury due to oxidative stress. We aimed to investigate the co-treatment effect of vitamin C as an antioxidant and platelet-richfibrin bioscaffold (PRF) as an angiogenic factor on reduction of ischemic-reperfusion injury in mice ovaries post autotransplantation. 30 female NMRI mice (18±2g) were divided into: control, autografted+normal saline (20 mg/kg/day), autografted+vitamin C (50 mg/kg/day), autografted+PRF and autografted+PRF+vitamin C groups and Treatment were carried out one day before until 7 days after transplantation. After 28 days, ovaries processed and stained with H&E method then analyzed stereologically and also the level of progesterone and estradiol hormones were measured. Data were analysed with SPSS software using one-way ANOVA and tukey’s test and the means differences was considered significant at p≤0.05. A significant reduction (p<0.05) in the mean total volume of ovary, volume of cortex and medulla as well as in estradiol and progesterone level was found in the autografted +normal saline group compared to the control. These parameters significantly increased in the autografted + vitamin C and autografted + vitamin C + PRF groups to the control level when compared to the autografted + normal saline group. The results showed that treatment with PRF and vitamin C alone as well as together could protect the grafted ovary from ischemia-reperfusion injury and improved the grafts structure and function.
