Investigation of thyroid hormones cell signaling in cardiac myocytes, with emphasis on the expression of 14.3.3 epsilon protein gene in male rats

Thyroid diseases is one of the most common diseases. Since the heart is one of the main target tissues for thyroid hormones, there are marked changes in the function of the heart in thyroid disease. So, determining biomarkers for thyroid diseases is important. 14.3.3 proteins are a group of highly conserved proteins that are involved in many cellular processes. All seven family members 14.3.3 are expressed in the heart. Here the effect of thyroid hormone on 14.3.3e expression in heart tissue was investigated. Samples were isolated from thirty Wistar adult male rat (weighing 250±50 g) that were divided into three groups control (intact), hypothyroid (6-Propyl-2-thiouracil (PTU) 0.05g/l for 8 weeks) and treatment (Levothyroxine 0.2g/l for 8 weeks). Total RAN was extracted by using RNXplus reagent, cDNA was synthesized by random hexamer primer RT reagent kit (Takara). Expression of 14.3.3e was evaluated by qRT-PCR. GAPDH was used as an internal control gene. The relative expression of 14.3.3e in hypothyroid and treated groups has not shown any significant changes (P> 0.05). Therefore, the hypothyroid induction in the cardiovascular system is not due to the effect of the hormone thyroid on the expression of the gene expression of this isoform and should consider other pathways. However, more studies with a high number of specimens are needed to make sure.
