چکیده مقاله:

It has been shown that curcumin has effective anti-cancer properties. However, the absorption efficacy of curcumin is too low to have significant results in therapy. To overcome this problem, we first improved the bioavailability of curcumin by making a Nano-based compound of Gemini Curcumin and then studied the effects of this compound on cell viability and apoptosis. Human gastric carcinoma AGS cell lines were treated with Gemini Curcumin (GC), and cell viability was evaluated using the MTT assay. Cell cycle analysis was assessed by flow cytometry and also the apoptotic potential of GC of was measured using Fluorescence microscope and Annexin-FITC/PI Apoptosis Assay. Expression levels of some genes involved in apoptosis pathways such as BAX and BCL2 were analyzed by qPCR and Western blotting. GC affected the cell viability of AGS cells in a dose- and time-dependent manner with IC50 values of 15 μM at 72 h of treatment. Our results showed that treatment with GC increases BAX/BCL2 ratio and induces significant G2/M cell cycle arrest in AGS cells. Gemini curcumin induces G2/M-phase arrest in human gastric cancer cells, which highlights its potent anti-cancer activity and potential application in gastric cancer therapy.
