Introduction & Aim: Colorectal cancer (CRC) is the third most common cancer and the fourth most common cause of cancer-related death worldwide. miRNAs are aberrantly expressed in cancer and influence tumor progression. The aim of this study was investigating the effect of sodium butyrate (NaBu) on miR-21 expression in HCT-116 cell line. Methods: HCT-116 Cell line was maintained in DMEM with 10% fetal bovine serum. Cells were seeded into 96-well plates, and then treated with various concentrations of NaBu (from 200mM to 6.25mM). After 24h, 48h and 72h incubation, the effect of NaBu treatment on the viability of cells was measured by MTT assay. RNA was extracted and cDNA was synthesized. qRT-PCR was done. Results: Our results showed that NaBu induced of cell death in a dose- and time-dependent manner. MTT assays showed that treatment with NaBu at high concentrations significantly inhibited the growth of HCT-116 cells. On the base of RT-PCR results, NaBu after 24 , 48 and 72 hours caused a significant up-regulation in the expression of the miR-21 in comparison with untreated cells as control group (P<0.05). Conclusion: These findings suggest that up-regulation of miR-21 gene expression is modulated by histone acetylation. This upregulation maybe effects on its target and regulated the expression it.
