Glucose can be covalen bond to the DNA as a non-enzymatic [1]. The persistence of DNA damage withincreasing agecould exacerbate the effects of glycation nucleotides [2].Due to the structural featuresKetone bodies acetoacetate and production free radical, the acetoacetatecan be effectivein the processbinding glucose to DNA and proteins [3].In the study the effect of acetoacetate on the AGEs formation ofDNA extractedwas studied under physiological conditions after incubation with glucose for 28 days,usingvarious techniques such as Circular dicroism (CD), gel electrophoresis,Fluorescenceand UV-visspectroscopy.The results of UV-vis spectroscopy show the amount of DNA-glycation in the presence ofacetoacetate increased. As well as its production of reactive oxygen species, increase the complications ofdiabetes. A new absorbance peaks appear between 300 and 400 nm, which indicate the formation ofDNA-AGEs. The modified DNA with glucose and acetoacetate had more absorbanceabout 0.5 thanmodified DNA with glucose alone. The DNA glycation in the presence of ketone bodies acetoacetateexacerbate structural changes in DNA, DNA single-strand break and increase of mutations and geneticinstability is caused.
