چکیده مقاله:

Among transition metal complexes palladium(II) complexes are very interesting candidates for alternativemetal-based drugs, because the coordination geometry and complex forming processes of palladium(II)are very similar to those of platinum(II)[1-3].Thus our research in this area is focused on the interaction ofCalf Thymus DNA (CT-DNA)with a new palladium(II) anticancer complex of[Pd(8Q)(phen)]NO3 (where8Q = 8-hydroxyquinolin and phen = 1,10-phenanthroline).There is a set of 4 binding sits(g) for thecomplex on the DNA with positive cooperativity in binding. n,the Hill coefficient find out to be 2.59 at300 K and 2.34 at 310K respectively. Kapp are 76.58 and 83.03 mM-1 at 300K and 310K respectively. Theabove compound can denature the DNA and the concentration of this ligand in the midpoint of transition([L]1/2), is decreased by improving temperature, from 0.057 at 300to 0.054mmol/L at 310K. the ΔG0H2Odetermined to be 47.63 and 46.68 kJ/mol at 300and 310K respectively. values for m, are 846.8and 881.3(kJ/mol).(mol/L)-1 at 300K and 310K respectively. ΔH0coformation in the range of 300K and 310K is find outto be 75.44 kJ/mol. ΔS0H2O of DNA denaturation by complex is 0.092 kJ/mol at 300K. Fluorescencetitration spectra and fluorescence Scatchard plots suggest that the Pd(II) complex intercalate in DNA. Thecytotoxicity assay of the complex has been performed on human breast adenocarcinoma MCF7 cell line,at micromolar concentration.
