Introduction: Lipases are glycerol ester hydrolyses that hydrolysis triacylglyceroles. This enzyme is water soluble and hydrolysis ester bonds in lipid substrates. Lipase is found in nature widely and produced by the most animal, plant, bacterial, yeast, and fungal species. Microbial lipases, specially thermo and alkaline tolerant, are respected greatly in biotechnology because of their resistance to high temperature and organic solvent in addition to catalysis ability of broad spectrum of bioreactions. They have large application in pharmaceutical, nutritious, washers, biodiesel and treatment of oil hog - washes.Material and Methods:The principle aim of this study is optimization of lipase production with response surface method by Natrialba asiatica. At the first step of experiment, lactose and Olive oil as carbon sources, peptone as a nitrogen source, sodium and magnesium ions, and environmental conditions such as pH, incubation temperature, incubation time and shaking rate were considered in 3 levels during the submerged fermentation by Plackett - Burman method. Despite of the importance of carbon and nitrogen resources in lipase production, the optimization with the method of Plackett - Burman showed that this resources have no effects on the production of lipase which made by Natrialba asiatica. At the second step, three parameters including incubation temperature, shaking rate and magnesium′s ion were considered in 5 levels used Central Composite Design.Results: The study showed that the best conditions for maximum production of lipase by Natrialba asiatica were 50°C, 133 rpm/min and 26 g/l of MgCl₂.Conclusion: The effect of incubation temperature and shaking rate were indicated that production of lipase by Natrialba asiatica is more dependent on physical conditions. As well as other lipases, magnesium′s ion had a positive effect on this matter.
