Response of carp (Cyprinus carpio) incubated ovarian follicles to stimulation by various exogenous hormones as measured by steroid secretion: A tissue culture approach

چکیده مقاله:

Objective: To study variation of steroid secretion by dispersed carp ovarian follicles as influenced by carp pituitary extract (CPE) and cultivated carp pituitary secretion (CCPS), Progesterone (P4) and estrogen (E2) levels in the culture media of carp ovarian follicles were estimated using radioimmunoassay (RIA). Method & Materials: Carp ovaries were collected and interstitial tissue was mechanically removed in order to obtain single follicles surrounded only by the follicular envelope. The ovarian follicles were cultivated in BSS Cortland medium alone or supplemented either with various amounts of CPE (50 and 100 μg/ml) or CCPS (50, 100 and 200 μg/ml. The culture media of ovarian follicles were collected after 72 hr and P4 and E2 levels were estimated using radioimmunoassay (RIA). In order to obtain CCPS carp pituitary primary cell culture was performed and the media culture collected after 72 hr and frozen at -20oc until use. Results & Conclusion: The highest and lowest concentrations of E2 and P4 were seen, respectively, in groups which influenced by 50 and 200 μg/ml CCPS. There was a significant increase of steroid secretion in group stimulated with 100 μg/ml CPE in comparison with the control group.
