Exhaustion Markers Expression on CD4 and CD8 T Cells due to Stimulation with Cell debris and Extracts of Food and Human-origin Probiotic Lactobacillus plantarum Strains

Background and Objectives:Probiotics exert various health-promoting functions of which immune regulatory activity is a strain specific character and an attention-seeking one. Immune exhaustion is a progressive loss of effector function of T cells due to prolonged antigen stimulation, characteristic of chronic infections and cancer. Probiotic strains might be effective in modulation of such immune responses. Materials and Methods:We aimed to assess the effect of wild strains of Lactobacillus (L)plantarum (PL4 and PT10 from food products; PF11 from human feces) with probiotic potential, on the expression of exhaustion markers (PD-1, Tim-3 and LAG-3) of T cells compared with standard probiotic strains L.plantarumATCC 14917 (PATCC) and L. acidophilus ATCC 4356 (AATCC).Peripheral blood mononuclear cells (PBMC) were treated with cell debris and cell extracts of the strains with and without Human anti CD3/CD28 T Cell Activator Beads, in order to determine immunomodulatory effects of the strains. Expression of the markers on CD4 and CD8 T cells was determined after 48hrs by flow cytometry and cell sorting techniques. Results: Although not statistically significant, most of the strains reduced the expression of exhaustion markers on CD4 and CD8 T cells except PL4 debris which significantly increased Tim-3 expression on CD8 T cells.When PBMC was pretreated with bacterial cell debris or extracts, was stimulated with anti CD3/CD28 beads, debris and extracts of strains PT10 and AATCC significantly reduced the expression of LAG-3 on CD4 T cells.Comparison between the strains showed that they didn’t exert any specific effect and just PL4 strain affected differently. Conclusion: Overall, expression of the above markers were less, when the cells were pretreated with bacterial cell extracts in comparison to debris. Strains PT10 and AATCC might be applicable in adjuvant therapy of chronic viral or cancerous diseases, which improve due to overexpression of exhaustion markers, after further examinations.
