Persian Golpar oils prevent acetaminophen induced liver injury through restoration of oxidative stress

Acetaminophen (APAP) is widely used as analgesic and antipyretic drug, but at overdose, it caused to undesirable side effects particularly hepatic necrosis. The objective of the present study was to investigate the hepatoprotective potential of H.persicum essential oil (E.O) against acetaminophen induced toxicity in rats. 80 male Wistar rats were randomly distributed into four groups (n=5); in negative control group (NC), the DMSO was only i.p injected. In control group (C), the cetaminophen (500 mg/kg b.w) dissolved in 400 μl DMSO was i.p injected. In the treatment groups, the H.persicumo E.O (100 and 200 mg/kg b.w) was diluted in 400 μl DMSO and injected i.p immediately after acetaminophen administration. Then, the degree of hepatoprotection was measured using antioxidant assays of liver homogenate including lipid peroxidation, reduced glutathione content, ferric reducing ability of plasma (LP, GSH & FRAP) at 4, 8, 16 and 24 h after acetaminophen treatment. Histopathological study of the rat liver was also done. The data indicated that the toxic effect of acetaminophen was controlled significantly by H. persicum E.O through restoration of the oxidative stress and antioxidant parameters; LP, GSH and FRAP towards near normal values. Furthermore, histology of the liver sections of the rats treated with the oils showed the presence of normal hepatic tissue in comparison with APAP group. These results strongly supported that H. persicum E.O extract could be an appropriate candidate for the treatment of acetaminophen induced oxidative stress liver damages.
