Tissue engineering approach using the combination of stem cells and nanofibrous scaffolds, has attractedthe interest of the research community for bone regeneration applications. In the present study, composed polymericnanofibrous polyethersulfone-polyethylene glycol (PES-PEG) produced by electrospinning were treated with mildNH3plasma to enhance cell attachment, growth, and differentiation. Prepared nanoscaffolds then was loaded withZn2SiO4 bioceramic nanoparticles. Scaffold Characterization was done by scanning electron microscope and contactangle measurements. The capacity of nanofibrous and loading Zn2SiO4 on nanofibrous scaffolds to supportproliferation and osteogenic differentiation of human Mesenchymal Stem Cells (hMSCs) was investigated using MTTassay and common osteogenic markers such as alkaline phosphatase (ALP) activity and calcium mineral deposition.The results of in vitro analysis showed that loading Zn2SiO4 on nanofibrous scaffolds significantly enhanced hMSCsproliferation and osteogenesis. Taken together, it was demonstrated that Zn2SiO4 on nanofibrous scaffolds aresuitable substrates to support the proliferation and osteogenic differentiation of hMSCs in vitro. They can also be anappropriate tissue-engineered matrix to support stem cells for bone tissue engineering.
