CALCIUM AND ALL-TRANS RETINOIC ACID DIMINISH THE DIFFERENTIATION OF HUMAN ADIPOSE-DERIVED STEM CELLS INTO ADIPOCYTES

Background and Aim: All-trans retinoic acid (ATRA) and calcium modulate the adipogenesis. In this study, the combination treatment of them on the differentiation of human Adipose-derived stem cells (hADSCs) to adipocytes was investigated. The hADSCs were differentiated into the adipocytes and simultaneously exposed to ATRA and calcium.Methods: The hADSCs positive markers; CD44, CD73, and CD90 and negative markers; CD31 and CD45 were measured by flow cytometry to ensure the purity of the cells. Besides, measurement of TG level, observation of lipid droplet by Oil Red O staining, GLUT4 concentration as a marker of adipocytes, and gene expression of PPAR-γ2, adiponectin, and GLUT4 was also evaluated by real-time PCRResults: While 10mM calcium did not change lipid accumulation, GLUT4 concentration, and the gene expression of PPAR-γ2, adiponectin, and GLUT4, these markers decreased in all treatment groups compared to the control group. There were no significant changes in gene and protein expression of GLUT4 in the treatment group of 0.5µM ATRA compared to the control. These markers significantly reduced in all combination treatments compared to the control. In comparison among the treatments groups, the best inhibitory result was seen in group 1µM ATRA+5mM calciumConclusion: These results suggest that the treatment of hADSCs with ATRA and calcium together can inhibit adipogenesis more significantly. Continuing with these studies may evaluate some helpful mechanisms that can be used in the prevention of obesity.
