Study the inhibitory effect of mesenchymal stem cells extracted from amniotic membrane (AM-MSC) on ovarian cancer (SK-OV-3 cell line)

IntroductionOvarian cancer is highest mortality rate among women s malignant tumors. MSC have immunomodulatory properties. Accumulating evidence suggests that mesenchymal stem cells are recruited to the tumor microenvironment. These cells can interact with cancer cells to promote invasion and metastasis and suppress tumor growth. Tumor microenvironment is an important actor of ovarian cancer progression but the relations between mesenchymal stem cells and ovarian cancer cells remain unclear. The objective of this study examines the inhibition effect of AMMSC on ovarian cancer.MethodsIn this study, we isolated MSCs from amniotic membraneand performed identity tests, then used ovarian cancer cell lines (SKOV3) and co-cultured them with MScs, and MTT assay was used to determine the proper inhibitory concentration of MSC on tumor cell. Then Interaction of MSC with Cancer Cells Using the co-culture Method. The Immunocytochemistry test was used to determine the expression of protein CDK2. Scratch test was used to determine the migration rate of cells. Cell cycle and apoptosis ware studying by expression of p53 and caspase3 genes by real time PCR respectively.ResultsAccording to the results MTT, Concentration 1:2 fold selected within 48 hours. The expression of CDK2 protein decreased. MSC reduced the migration of tumor cells. The level of expression of p53 and caspase3 was higher than control cells.The study showed that MSCs isolated from the amniotic membrane play a role in reducing the proliferation and migration and inducing apoptosis of ovarian cancer cells. This is done by increasing the expression of p53 and caspase3 genes and decreasing the expression of the CDK2 protein. So they are a proper cell for this purpose.
