IntroductionHydroxyapatite (HA) has been successfully coated on ZrO2 to construct a porous scaffold with higher mechanical strength but there are still big challenges regarding the delivery of osteogenic growth factors, local reserve, and sustained release of them. ObjectivesIn this study, we fabricated a porous interconnected HA/ZrO2/ Eggshell Membrane(ESM)/ platelet rich plasma (PRP) scaffold, compositions exhibiting anti-inflammatory, Osteoinductive and Osteoconductive activity and rich source of growth factors used in regeneration of tissues. MethodsPorous interconnected HA/ZrO2 scaffold were fabricated by slurry method with 71.6 + 0.5% porosity and 310 + 150 μm pore size. 10 μg/ml of Eggshell Membrane (ESM), were mixed with PRP as evenly spread. After activating PRP with 2.5%CaCl2, PRP/EMS followed by impregnation into the porous scaffold.ResultsCulturing MG63 cell line showed that 100 percent of cells attached to the PRP-containing scaffolds compared with HA/ZrO2 ones. The cells also showed a significantly higher alkaline phosphatase activity and calcium mineralization in scaffolds treated with PRP/ESM. MTT viability test demonstrated a significantly higher proliferation rate on the scaffolds containing PRP/ESM compared to those without PRP/ESM. ConclusionThe scaffolds with PRP/ESM seem to provide a superior microenvironment for osteoblast activities and can be suggested as a good vehicle for growth factor and cell delivery in bone tissue engineering applications
