IntroductionPlatelet rich plasma (PRP) has been recently used widely for many therapeutic purposes and tissue engineering applications. PRP has been used in combination with natural or synthetic biomaterials to fabricate engineered scaffolds for different purposes. SiO2 and platelet rich plasma (PRP) are two biomaterials that have profound impacts on ossification.ObjectivesThe objective of this study was to fabricate a SiO2/PRP composite to be used in bone repair.MethodsHuman PRP was prepared by centrifugation, mixed with SiO2 and induced to form a gel by CaCl2 supplementation. The gel exposed to glutaraldehyde for increasing gel stability and strength. FTIR test was performed to show any chemical change in the scaffold ingredients. Scanning electron microscopy (SEM), EDS and ICP was (were) performed to characterize the fabricated scaffolds. MG63 cell line was cultured on the scaffolds and cell viability was evaluated by MTT assay. The osteoblast functions were also evaluated by alkaline phosphatase activity assessment and alizarin red.ResultsSEM micrographs and EDS showed SiO2 particles were trapped within fibrin fibers in both crosslinked and non-crosslinked scaffolds. ICP revealed that crosslinking procedure did not influence the releasing amount of SiO2 into the medium. SiO2 containing scaffolds were provided a superior environment for cell proliferation and osteoblast activity.ConclusionBoth PRP and SiO2 provided a beneficial condition for the osteoblasts viability, proliferation and function. PRP/SiO2 scaffold can be considered as an autologous vehicle with appropriate features for bone tissue engineering
