چکیده مقاله:

Allograft organ transplantation remain the gold standard for the treatment of end-stage liver diseases. However, various issues, including donor shortage and need to using immunosuppressive drugs after surgery boasting demands for alternative solution. Recent advancements in tissue engineering field using bioprinting approaches, have enabled researchers to develop a novel 3D constructs that could mimic the functional unit of liver in some aspects including albumin secretion. But, total gene and proteins expression profiles significantly differ between these tissue engineered constructs and native human liver tissue. Important aspect of common bioprinting technique is a bioink. Scaffold based bioinks face significant challenges such as biocompatibility, immunogenicity and printability, which hinder the translation of this bench research to clinics. Cell sheet engineering strategies hold great promise in tissue engineering due to mimicking the developmental process of embryo. Application of cell sheet as a scaffold free bioink offer advantages due to lack of unprintable materials and can provide cell-cell interactions which increase ECM secretion by cells. Produced ECM act as a support for more cell proliferation, differentiation and viability in long term. Thus, continuous progress of cell sheet based bioinks may contribute to creation of novel 3D liver constructs
