Background: Chitosan is nontoxic, biocompatible and biodegradable, and with low antimicrobial activities. Here, the antibacterial activity of a modified chitosan (chitosan/ poly ethylene oxide) is tested through standard antimicrobial tests. Methods: The in vitro antibacterial activities of chitosan / poly ethylene oxide (Cs/Peo) were carried out using one gram-positive bacteria (Staphylococcus aureus) and one gram-negative bacteria (Escherichia coli). Practically, a micro-dilution technique was developed using 96-well microplates and tetrazolium salts to indicate bacterial growth. Microplate/p-lodonitrotetrazolium violet (INT) technique was applied to determine the minimum inhibitory concentration (MIC) of chitosan derivatives. All detections were repeated in triplicate. Results: Importantly, chitosan / poly ethylene oxide showed antibacterial activities against S. aureus and E. coli, Furthermore, chitosan / poly ethylene oxide showed broad and antibacterial activities. The MIC of chitosan / poly ethylene oxide ranged between 6.25 - 200 mg/L against both bacteria. Conclusion: It was found that chitosan / poly ethylene oxide showed antibacterial activities against both tested Bacteria. The broad and high antibacterial activities of chitosan / poly ethylene oxide might be related with the grafted–NH2 on the chitosan molecule.
