Background and objective: Mesenchymal stem cells (MSCs), is a multipotent adult stem cells that have been isolated from several adult tissue sources, such as, adipose tissue, which named as Adipose-derived Stem Cells (ASCs). In the past decade, adipose tissue has become a greatly interesting source of adult stem cells with the multipotent ability such as, immune sickness treatment and regenerative medicine and etc. The isolation of Adipose-derived Stem Cells currently is based on the use of the enzymatic system, the Collagenase that consumes in this method is an expensive reagent derived from a bacterial source. On the other hand today there are thousands of articles now describe the use of ASCs in a variety of therapeutic applications of ASCs. Therefore to ensure the highest possible safety and more efficient performance and more quality of the cells are isolated and lower prices and easier access, a precisely defined system with a better-quality control is required. Our experiment evaluated the extraction of mesenchymal stem cell from lipoaspirate tissue lacking an enzymatic digest. Materials and method: Here, we used a simpler and cheaper method called cellular escape or non-enzymatic systems from adipose tissue to collect and isolate the stem cells. For this method, after washing the liposuction sample, Place the tissue pieces in dishes containing a complete culture medium, and lets they stayed in the incubator without moving for few days, then Stem cells will be extracted by the cell scape method because , there is a richer environment in medium than tissue pieces. And at the end, the results were compared with matched non-enzymatic systems with the enzymatic method by collagenase, based on stem cell yield and differentiation potential. Findings: The results revealed the usability of this method for stem cell isolation from Adipose tissue and indicated the same differentiation potential and profile compared to the collagenase system. These results are in terms of adipogenic and osteogenic differentiation potential. Conclusions: Consequently, the two approaches yield the same cell product. Although using enzymatic system increases cell yield.
