چکیده مقاله:

Colon cancer is the third most common cancers in the world which causing harms & wastecost. In the present study we investigated changing of mechanical properties of cells andtheir effects on bio physic chemical signals to determine injuries factors and betterunderstanding of metastasis process. Ht29 Cancer Cell line was purchased from Pastureinstitute. The cells were incubated with 1640RPMI media (Sigma) & 10%FBS serum (sigma)over a period of 3 days. Experimental cell group treated by albendazole (200 Mol.) andcompared by control group. The amount of Cell microtubules as a major factor in mechanicalproperties was accessed by staining with polidine florescence isotiosianate (50mg/ml)(sigma) & Anti β tubuline CY3 (sigma) and finally was studied by a florescence reversemicroscope. Furthermore the cell groups were studied by staining with MTT assay (ELISA)reader. The results showed significant decreased in tubulins of cells after treatment .so actinratio to microtubules increased and loss of fluorescence reactivity in cells. These cellsseemed lose their mechanical properties in compare of amount of control cells. The resultsshowed the using of albendazole caused a reduction in the number of the cells. Thesefindings may suggest more attention to apply methods for altering and control of the cancercell behaviors for movement , diapedesis and metastasis process as a new strategy fortreatment of cancer.
