چکیده مقاله:

Cancer is a kind of genetic disease which eventuate in an abnormally division of cells in anuncontrolled manner. Cells in malignant tumours can invade nearby tissues and spread toother parts of the body. According to the World Health Organization, breast cancer is the mostcommon diagnosed cancer in women. MicroRNAs are small non-coding RNAs that playimportant role in gene expression and molecular regulation. They can target importantmRNAs which act in different signaling pathways such as cell cycle, cell migration, apoptosisand angiogenesis for cleavage or translational repression. mRNA targetome of microRNAscan be investigated to assess the oncogenic or tumor suppressive role of microRNAs. In thisstudy, we analyse miR-499-5p targetome in breast cancer. The aim of this study is toevaluate molecular role of miR-499-5p as a potential biomarker in breast tumor cells by usingbioinformatics tools. Validated and predicted targets of miR-499-5p were obtained frommiRtarbase and miRwalk database, then DAVID database was used for miR-499-5penrichment analysis. To determine gene expression of its targetome in mammary glands andbreast tissues, uniGene database was used. There are about 1815 predicted miR-499-5ptargets in KEGG signaling pathways. Among them, 47genes are related to cancer pathwaysand also more than 27 genes are expressed in mammary gland tissues and breast tumor. This is predicted that some important cancer-related mRNAs such as KITLG, FGF, PDGFR,Bcl2 ,STAT3 are targeted by miR-499-5p. According to our data, miR-499-5p may be relatedto breast cancer through targeting mRNAs in different vital signaling pathways. It has beenindicated that miR-499-5p targetome such as KITLG, FGF, PDGFR, Bcl2, STAT3 leading toproliferation, evading apoptosis and sustained angiogenesis in breast cancer. Therefore miR-499-5p can be used as a proper prognostic biomarker.
