Recently, the Schiff base compounds have attracted much attention because of biologicalcharacteristics such as antioxidant, antibacterial, anti-inflammatory and antitumor properties.Studies show that complexes of schiff base containing copper, are able to inhibit the growthof cancer cells by inhibiting proteasome activity. Ubiquitin-proteasome pathway is involved inthe regulation of cell death and proliferation. In this study, were investigated the antiproliferativeand antioxidant properties of Cu2+ complex of the N,N′-dipyridoxyl(1,2-diaminobenzene) schiff base on breast cancer cells (MCF7). MCF7 cells were cultivated andtreated with different concentrations of Cu2+ complex of the N,N′-dipyridoxyl(1,2-diaminobenzene) schiff base (0, 125, 250, 500, 1000 μg/ml). ÐoeТТ assay was done forexamined the anti-proliferative effects of schiff base and the occurrence of cell death wasassessed by changes in morphology of cells. Antioxidant properties of this schiff base wasevaluated by scavenging of 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical. The results ofMTT assay showed that the cell proliferation was decreased as time and dose dependentmanner with IC50 about 500 Âμg/ml. Morphological analysis reveals changes in treated cellssuch as decrease volume of cells, changes in the shape (cell shrinkage and cytoplasmicprotrusions) and size of cells. Furthermore this schiff base acted as 1,1-diphenyl-2-picrylhydrazyl (DPPH) (IC50=300Âμg/ml), radical scavenger. The results of this studysuggest that complexes of schiff base containing copper have the potential to become an anticanceragent.
