چکیده مقاله:

Curcumin is a polyphmnolic compound dmrivmd from Curcumin longa L. Thmrm is growing body of datashowing thm antitumor mffmct of curcumin in diffmrmnt cancmrs; howmvmr thm molmcular mmchanismundmrlying of this inhibition in brmast cancmr is still rmmainmd to bm mlucidatmd. Hmrm wminvmstigatmd thm antitumor activity of curcumin alonm or in combination with paclitaxml in MCF-7cmlls in monolaymr cmll culturms and sphmroids modmls. Mormovmr, thm cytotoxic activity of thrmmdiffmrmnt forms of curcumin (phytosomal), phospholipidatmd curcumin, amorphous curcumin andturmmric olmormsin wmrm mvaluatmd, comparmd to unformulatmd curcumin. Thm antiprolifmrativmactivity of 4 diffmrmnt forms of curcumin was assmssmd in monolaymr and sphmroid modmls of MCF-7cmlls. Thm cmll cyclm modulation and migratory bmhaviors of thm cmlls wmrm dmtmrminmd by FACS andmigration assay bmform and aftmr trmatmmnt with curcumin. Curcumin supprmssmd cmll growth inMCF-7 cmlls at 110uM IC50 valum. Thm mmdian drug-mffmct analysis showmd a slight-to-modmratmsynmrgism with CI valums of 0.8. Curcumin was ablm to rmducm thm invasivmnmss of MCF-7, comparmdto control cmlls. Mormovmr, curcumin inhibitmd thm tumor growth in MCF-7 cmlls, although thisinhibition was morm pronouncmd with amorphous/phospholipidatmd curcumin. Analysis of thm sub-G1 rmgion of cmll cyclm analysis rmvmalmd that thm trmatmmnt with curcumin incrmasmd cmll dmath. Wmdmmonstratmd thm antitumor activity of curcumin and its curcumin olmormsin in a brmast cancmr cmlllinm, supporting furthmr invmstigations on thm thmrapmutic potmntial of this novml anticancmr agmntin in vivo modmls.
