Cd4+ memory subsets in tumor draining lymph nodes of bladder cancer

The compartments of memory T cells play a fundamental role in the immune system by substantiating specific and acquired immunity. Subsets of memory T cells are heterogeneous and could be divided into various subtypes based on the expression of surface markers and their functionality. In the present study, we aimed to determine the frequency of CD4+ T memory cell subsets in tumor draining lymphnodes of bladder cancer (BLC) patients. Mononuclear cells were obtained from lymph nodes of 45 untreated patients with BLC and stained with anti-CD4, -CCR7, -CD45RO and -CD95 antibodies. Data were acquired on a four-color FACSCalibur flow cytometer and analyzed using CellQuest Pro software. Although no significant association was found between CD4+ memory subsets and most of clinic-pathological parameters, total frequency of effector memory T cells (CD4+CCR7- CD45RO+CD95+) was significantly higher in tumor-negative lymph nodes compared to tumor positive ones (p> 0.05). The frequency of these cells (p=0.032) also significantly increased in patients with low histological grade tumor comparing to high grade ones. The increase in the percentage of CD4+ effector memory T cells in tumor free lymph nodes as well as patients with lower grade tumors could imply a protective role for these cells in preventing tumor metastasis to lymph nodes
