Cytotoxic effects of biosynthesized zinc oxide nanoparticles biosynthesized with origanum majorana extracts on mda-mb-231 breast cancer cell line

Among the metal oxide nanoparticles, Zinc Oxide nanoparticles (ZnO-NPs) is investigated to be one of the best exploited at Nano dimensions. ZnO-NPs is a wide band-gap semiconductor, which has been applied in many fields such as pharmaceutics, cosmetic, the fabrication of solar cells, photocatalysis, gas sensors, coatings and biomedical. The purpose of this study was to examine the antioxidant and cytotoxic effects of ZnO-NPs biosynthesized with Marjoram extract on MDA-MB-231 breast cancer cells. For MTT assay, the MDA-mb231 cells were cultured (1x105 cells/ml) in a special culture plate. Then, the cancerous cells were treated with different doses of ZnO-NPs/M for 24 hr. After the incubation time, the survival rate was estimated by MTT assay.DPPH radical scavenging assay was done for evaluating the ability of nanoparticle to trap the DPPH free radicals. 1 mg of DPPH was dissolved in 96% ethanol to produce DPPH free radicals. Stock solution ZnO-NPs/M of was ready using the solvent in various doses (0-500 μg/ml). The reaction mixture was contained 500μl of ml DPPH solution and 500μl ZnO-NPs/M. Absorption was measured at 517 nm wavelength 30 minutes after incubation.The data of the MTT procedure exhibited that ZnO-NPs/M reduced the viability of MDA-MB-231cells in dose-dependently. The IC50 during incubation for 48 hours was determined 60 μg/mL (P<0.05). The ZnO-NPs/M exhibited a concentration-dependent activity and also the DPPH scavenging capacity was 43.4 % at a concentration of 500 μg/.Zinc oxide nanoparticles biosynthesized with Marjoram water extract have the antioxidant and cytotoxic effect and they can be used in biomedical application specially for the treatment of breast cancer.
