Fabrication and characterization of micelles incorporating Hydrocortisone as avehicle of drug delivery

چکیده مقاله:

Background and objective: Hydrocortisone is a glucocorticoid that it’s pharmacological actions are Immunosuppressive, Vasoconstrictive, Anti inflammatory and Anti proliferative effects[1]. It has been used extensively in managing many oral diseases, due to its excellent anti-inflammatory and immuno-modulatory effects[2]. Some of the indications of Hydrocortisone in dentistry are in RAS(Recurrent Aphthous Stomatitis), Oral lichen planus, Bullous pemphigoid, restorative dentistry and so on. It is employed in the topical forms including ointment, paste, mouthwash and systemic forms.Poor water solubility is the major defect of this drug that causes manifestation of side effects. The polymeric self-assembled nanoparticles offers some unique advantages including core-shell morphology, high loading capacity, site-specific drug delivery, and avoids unwanted side effects of administered drug. Moreover, micelles remain stable in blood circulation for prolonged period of time [3]. Because of this reasons, Loading of Hydrocortisone into Dextran-PLGA micelles can decrease its side effects.Materials and methods: Dextran-PLGA copolymer incorporating Hydrocortisone was synthesized to make polymeric micelles for drug delivery. Physicochemical properties of fabricated system evaluated with FT-IR, NMR and TEM and critical micelle concentration of fabricated system determined.Findings: Polymeric micelles was spherical and smaller than 300 nm with appropriate size distribution. Chemical bonding of Dextran-PLGA confirmed with FT-IR and NMR. Critical micelle concentration of polymer for micelles was 50 g/ml.Conclusion: By synthesis the micelles composed of Dextran-PLGA copolymer and incorporating Hydrocortisone into them, it is expected that Hydrocortisone bonds to hydrophobic PLGA in core of the micelle and Dextran as a hydrophilic polymer on the surface of the micelle leads to dissolution of micelle in body’s fluids and causes the micelles to pass the reticuloendothelial systems. This model of drug delivery causes to decrease drug side effects and better compliance of patients and can be used in many dental problems
