Dracocephalum Kotschyi L. is an important rare-medicinal plant which belongs to the Lamiaceae familly. Recently, much attention has paid to Dracocephalum genus and its chemical constituents because of their diverse activities, such as Anticancer, antioxidant, anti hypoxic and immunomodulatory activities [1]. Production of this plant is limited due to seed dormancy and several biotic stresses of which wilt disease is the most serious. In vitro culture techniquesrepresents an excellent option for the mass-propagation and conservation of rare, threatened or endangered medicinal plants [2] The main objective of this study is to achieve a suitable method for in vitro regeneration of D. Kotschyi via nodal cutting technique.MATERIALS AND METHODS Five Nodal segments of D. Kotschyi derived from in vitro germinated seeds were cultured per bottle containing MS media including various concentration of Zea (0. 1 and 2 mg/l) combined with IBA or NAA at 0, 0.2 and 0.5 mg/l. Experiment was set up in completely randomized block design with6 replications (bottles). Data were recorded on parameter after 4 weeks and were analyzed using theSAS version 9.1 statistical computer program. When the ANOVA indicated significant treatment effects (5 or 1%) based on the F-test, the Duncan’s multiple range test (P<0.5) was used todetermine which treatments were significantly different from other treatments. RESULTS AND DISCUSSIONPlant growth regulators had highly significant effects on the measured parameters (Data were not shown). The highest percentages of the length of the stem 1.2 cm and number of leaf and bud 1.6 cm were observed on MS medium containing 2 mg/l ZEA+0.5 mg/l IBA. Regeneratred shoots were rooted on the same medium then transferred to greenhouse for further growth and development. Finally the acclimated plants were then shifted to the field condi-tions having 100% survived. This These results confirmed by [3] reported that high frequency and rapid regeneration protocol from shoot tip and nodal explants of Menthe piperta L. has taken place on (MS) supplemented with either BAP (1Mg/l) or Zea (0.25mg/l).
