Antimicrobial Activity Of Silver Nanoparticles Is Importance To Control Conventional Antimicrobial Agents. Bacterial Contamination Is A Serious Problem In Plant Tissue Culture Procedures. The Concentration Of Tio2 Usually Required To Kill Bacteria Varies And Depending On The Size Of The Particles And The Intensity And Wavelength Of The Light Used. The Antibacterial Activity Of Tio2 Has Been Found To Be Due To A Reaction Of The Tio2 Surface With Water. This Research Was Planning To Evaluate The Potential Of Nano Silver (NS) And Titanium Dioxide Nanoparticles (Tio2) To Remove Bacterial Contaminants.Materials and methods:In this research elected tobacco (Nicotinia tabacum cv. Xanthi) from Biotechnology department of Isfahan University of Technology and propagated in MS medium through bud cultivating. We added different amount of nano silver and TiO2 to tissue culture media and the explants were placed on Murashige and Skoog (MS) media. Experiment involved TiO2 NPs with 2 rates (1 and 2 w/w) in MS medium. Experiment involved NS with five rates (8, 10, 20, 50, 80 mg L-1) in MS medium.We evaluate the explants after four weeks.The percentages of nano materials antimicrobial activity that used in tissue culture media were recorded four weeks after culture. The experiment was conducted as a completely randomized design in a factorial arrangement with three replications.Result and discussion:Successful tissue culture of all plants depends on the removal of exogenous and endogenous contaminating microorganisms. Fungi and bacteria are the most common microorganisms to be found on or in plant tissues. Several nanomaterials, such as NS, and TiO2, have found applications in diverse consumer products and industrial processes, including water treatment, as antimicrobial agents. In this research we study the antimicrobial activity of nano silver and nano dioxide titanium. After confirm the ability of nano silver and nano dioxide titanium to reduce the microorganism, we decide to using and adding NS and TiO2 to MS tissue culture media. The results show that NS and TiO2 can reduce and remove microorganisms in MS media and then the explants can growth very well. It can be suggested that the TiO2 nanoparticles and NS may be a useful material for removing the bacterial contaminants in plant tissue culture.
