چکیده مقاله:
Transdermal systems are among Novel Drug Delivery Systems and are attributed to the systems having property of diffusing drugs through skin in a noninvasive form by delivering drugs into systemic blood circulation which as a consequence the drug effects appears. Constant blood level, non-hepatic first pass effect, patience compliance are advantages of these systems compare to oral administration and intravenous infusion. The present research was designed to evaluate different new Iranian drug-in-adhesive formulations of nitroglycerin transdermal patches for the treatment of angina pectoris. The target was to design drug-in-adhesive patches (DIAPs); a full factorial design was used. Different types and amounts of pressure- sensitive adhesives (PSAs), chemical permeation enhancers (CPEs) were used and evaluated with respect to drug release
and adhesive properties. A simple but precise method, the peel 180° test and Probe Tack Test, was developed to measure and compare adhesive properties of transdermal patches. Addition of CPEs was led to decline of the mechanical properties of PSA, which can produce some difficulties in manufacturing of transdermal systems. In-Vitro release profiles of nitroglycerin from different formulations depended on viscosity and
type of pressure sensitive adhesive and type and concentrati on of selected chemical permeation enhancer. The results showed that release kinetics obeyed the square root of time or Higuchi model, indicating the diffusion controlled release mechanism. It was found that the amount of nitroglycerin needed for each 15 cm2 one-day DIAP should be 40 mg. It was concluded that acrylic PSAs showed good adhesion and release
properties.
