Background and Aim: Effectiveness of MSCs in the treatment of wideranges of disease (e.g., hematopoietic recovery, inflammatory diseases,bone regeneration, infarcted myocardium, joint diseases etc) has beenwell documented. As a result, isolation and characterization of MSCsfrom different species for extensive preclinical studies are required.Zebrafish liver tissue-derived MSCs described here fulfill the basic MSCcharacteristics and these criteria. The aim of this study was to investigatethe immunophenotypical characterization of liver-derived MSCs for thefirst time for using zebrafish as a suitable animal model for regenerativemedicine in cell therapy.Methods: In this experimental study, zebrafish (Danio rerio) waspurchased from a local fish dealer. Liver of zebrafish was obtainedand carefully minced under sterile conditions. In the following, MSCswere isolated enzymatically from liver tissue of Zebrafish as previouslyreported in another study by Fathi et al. Subsequently, the RT-PCR, aswell as flow-cytometry, were used to analyze the expression of a panelof immunological cell surface markers CD44, CD90, CD31, and CD34.Also, alizarin red, oil Red-O, and toluidine blue staining were carriedout to evaluate the multipotency of zebrafish liver tissue-derived MSCs.Results: In the immunophenotyping analysis, the liver-derived MSCswas found to be immunopositive for CD44 and CD90 epitopes asmesenchymal stem cell markers, and immunonegative for CD31 andCD34 epitopes as hematopoietic stem cell markers. These results are, incombination with the RT-PCR results, were also positive and negative forgene expression of CD44 and CD90 and CD31 and CD34, respectively.These findings are in line with the results of other MSCs characterization;these reactions are generally characteristic of MSCs.Conclusion: Our results show that liver tissue-derived MSCs exhibitedexpression of a typical set of classic MSC surface markers, which willcontribute to a more extensive characterization of zebrafish MSCs priorto their use in cell therapy and regenerative medicine.
