Background and Aim: Human mesenchymal stem cells (MSCs) havebeen of recent particular interest due to their unique features of selfrenewaland potency for differentiation to many somatic cell types whichis the basis for regenerative medicine. The parp1 plays an indispensablerole in the preservation of this process because it can act together with other pluripotency factors. RNAi strategy including siRNA and miRNArepresents a post-translational gene expression modification. miRNAbasedtherapies face with some obstacles such as difficulty in passing thecell membrane barrier and also passage within the cells.Methods: In the present study, a new liposome-based formulationincorporating two miRNAs was synthesized and characterized by precisetechniques in order to confirm its optimization in terms of stability andefficient transfection. Then free miRNA and lipoplexes containing miRNAwere used for transfection into MSCs, and their effect on the expressionlevel of parp1 was evaluated by qPCR.Results: Parp1 gene expression was over-expressed in the presence ofmiR-302a, however, miR-34a had the opposite effect on Parp1 mRNAlevels and down-regulated the level of Parp1 mRNA significantly.Conclusion: Our findings suggest that parp1 is a key component in theregulation of pluripotent cells and this can be regulated by the use ofmiRNAs. The high efficiency of the novel formulation of the liposome ispromising.
