Background and Objective: As microbial resistance against antibiotics is increasingly growing, it is necessary to develop new remedies. Silver nanoparticles have shown significant antibacterial properties. Synthesis of silver nanoparticles mediated by plant extracts could be considered as an alternative in treatment of various infections. The objective of this study is to find out the antimicrobial effect of Scrophularia striata Boiss. aerial organ extract silver nanoparticles Materials and Methods: In this study silver nanoparticles have been synthesized using Scrophularia striata Boiss. aerial organ extract. Optimum temperature, concentration and also optimum pH for production of silver nanoparticles were established. Formation of the particles has been investigated by UV-vis spectra, EDS, XRD and SEM techniques. The antibacterial effect of these particles then has been studied. Ciprofloxacin was used as positive control. Determination of minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were carried out. Five gram-negative bacteria strains such as Acinetobacter baumannii (ATCC 19606) and two gram-positive bacteria strains such as Staphylococcus aureus (ATCC 29213) were used in the study. Findings: The optimum condition of this green synthesis was 85°c, best pH 11, optimum concentration of silver nitrate was 8 mM and 30 minutes was the best time for the synthesis. These nanoparticles made by green synthesis from methanolic extract of Scrophularia striata Boiss. aerial organ extract showed the highest antimicrobial activity for Staphylococcus aureus (MIC= 1.2μg/mL, MBC= 37.5μg/mL) in compared to Ciprofloxacin (MIC=0.21μg/mL). Ciprofloxacin was used as a positive control in the MIC and MBC test. The average size of nanoparticles was 40 nm showed by the SEM test. Conclusion: According to the results of this study, silver nanoparticles made from methanolic extract of Scrophularia striata Boiss. aerial organ extract synthesizes by green synthesis method showed very good antimicrobial activity and can be an alternative product for antibiotics.
