Impact of extraction methods on antioxidant activity of Oxalis corniculata, evaluation of antihypoxic activity, green synthesis of silver nanoparticles and evaluation of their antimicrobial activities

Background: Oxalis corniculata is a well-known medicinal plant, abundantly distributed in Guilan, with a wide spectrum of biological activity. This investigation was design to study the impact of extraction methods on its total phenolic contents, antioxidant and antihypoxic properties. The extract was used for preparation of Ag nanoparticles (AgNPs). Method: Aerial parts were extracted by meceration method, ultrasonic assisted (UAE) and soxhlet assisted extraction (SAE). Antioxidative capacity was assessed by DPPH and nitric oxide (NO) free radicals scavenging, reducing power and iron chelating activity. Total phenolic and flavonoid contents were determined. Antihypoxic activities of extract against hypoxia-induced lethality in mice were evaluated by asphyctic, haemic and circulatory models.The extract was used for preparation of AgNPs. Optimum conditions were established and finally the antibacterial effects of AgNPs were studied against 7 bacteria. Results: UAE showed the highest amount of total phenolics and total flavonoid contents. In DPPH radical scavenging activity, UAE had a higher activity which was significantly different from other extracts. Extraction method did not show any effect on reducing power, extracts showed the same activity (P> 0.05). SAE show the best NO radical scavenging activities. UAE showed the best iron chelating activities. In haemic model, extract at 125 mg/kg showed statistically significant activitiesrespect to the control (p<0.01). Extract significantly prolonged the latency fordeath with respect to control group. In circulatory and asphytic models, extract at 250 mg/kg, significantly prolonged the latency for death with respect to control groups (p<0.001).Formation of AgNPs were confirmed by UV-vis spectra, EDS, XRD and SEM techniques. Determination of MIC and MBC revealed that AgNPs shows strong antibacterial effects on P. aeruginosa, and high antibacterial activities against S. aureus and E. coi. Conclusion: The results clearly indicate that extraction methods significantly affect antioxidant capacities and total phenolic contents. For this plant, UAE was chosen as more efficient method for extraction of antioxidants. Extract showed strong antihypoxic properties. Extract had high potential for preparation of AgNPs.
