Residue analysis of Sulfonylurea pesticides in wheat using liquid chromatography–tandem mass spectrometry

Objectives: Sulfonylurea pesticides are new classes of herbicides, used as selective pre and post-emergence herbicides for control of most weeds and annual grasses. This class pesticides containing a homogeneous group of the general formula R1- NH-O=C-NH-SO2-R2 (−R3), where R3 ortho substitution on the ring. This study represent, a liquid chromatography–tandem mass spectrometry (LC-MS/MS) method for the analysis of sulfonylurea pesticide residues in cereal which was developed, optimized, and successfully performed for analyzing routine samples in food reference control laboratory.Methods: The samples were extracted with acetonitrile containing 1% formic acid, and salted out with MgSO4, NaCl, and citrate buffer without extra cleaned up procedure. The sulfonylurea pesticides were analyzed by negative electrospray ionization in Multiple Reaction Monitoring mode. Sulfonylurea pesticides were separated on a reversed-phase C18 column Chromolith performance RP-18e 100-4.6mm with a gradient elution using water-acetonitrile with%0.01 acetic acid mobile phase at a flow rate of 0.4 mL/min. Quantification of the sulfonylureas were obtained using spiked calibration curve on the wheat matrix.Results: The linearity of pesticides was investigated at five concentration level (10-200 ng g-1) and also, the recovery of pesticides was evaluated at three concentration levels 15, 75, and 120 ng g-1 (n=3).The correlation coefficients (r2) were higher than 0.990.The repeatability (in term of RSD%) of the method was obtained for pesticides in the range of 0.5-20% in wheat matrix. The limits of quantification and detection for most of pesticides were 10 ngg-1and 3.3 ngg-1, respectively.Conclusion: The performed method with using LC-MS/MS is sufficiently accurate, precise and sensitive for analyzing Sulfonylurea pesticides in cereals in monitoring and control program.
