Evaluation the effect of main constituent of Lippia citriodora Leaves, on paraquat-induced toxicity in-vitro

Purpose: Paraquat (PQ) is a widely used herbicide. PQ exposure in acute and chronic may cause a range of side effects from gastrointestinal problems to fibrosis and renal failure. Treatment for PQ toxicity consists primarily of supportive care and there is no FDA- approved treatment. In the present study, we aimed to elucidate the effects of verbascoside as phenyl propanoid compound with antioxidant and anti inflamtory properties, on PQ-induced lung toxicity in A549 cell line.Methods: A549 cells were exposed to 12.5 to 50 μM concentrations of verbascoside and 300μM PQ and cell viability was measured using MTT assay after 24 hours of exposure. Intracellular ROS production was measured by fluorometric using 2 , 7’-Dichlorofluorescin diacetate (DCF-DA). The activity of enzymes including super oxid dismutase, catalase and gluthathion peroxidase were evaluated using ELIZA kit. In addition, the level of NF-KB was also assessed using the western blot analysis.Results: The result of this study showed that pretreatment of A549 cells with verbascoside (12.5, 25 and 50 μM) and then PQ (300μM) could reduce the PQ-induced cell cytotoicity in A549 cell line. PQ exposure increased the amount of ROS in comparison with control group that was reversed by verbascoside admintration in a concentration dependent manner. Indeed, the concentration of 25 and 50 μM VB +PQ, increased the activity level of SOD, catalase and GPx compared to group that received PQ alone. The results also show a significant decrease of the expression of NF-KB when the cells were treated with VB +PQ.Conclusion: Our findings might raise a possibility of potential therapeutic application of verbascoside, for preventing and treating PQ-induced lung toxicity. Evaluation the effect of VB on other adverse effects of PQ, is recommended.
