Introduction: The COMT gene is on chromosome 22 and involves in estrogen metabolism. The polymorphic of this gene can affect its expression and contribute to the development of cancer. In this study, the relationship between rs2075507 polymorphism in the promoter region of genes was investigated in breast cancer of Iranian women. Methods: Blood samples were received from 100 breast cancer patients and 100 healthy individuals from Shohada Tajrish Hospital of Tehran. After extracting the DNA and examining the quality and quantity of them, the specific primers designed for replication. All samples were replicated and digested with a specific enzyme. The results of enzymatic digestion, used for genotyping and results were evaluated by chi-square and logistic regression. Results: The mutant genotype GG did not show a statistically significant relationship with breast cancer (p.value: 0.7, 0.95% CI: 0.544-2.475), and its odds ratio was 1.161, indicating the effect of genetic variation on infection Breast cancer. The wild genotype AA have showed statistically significant relationship with breast cancer (p.value: 0.19, 95%CI: 0.251-0.888), but the odds ratio of was showen 0.473 a decreasing effect on the risk of breast cancer, and there was no statistically significant relationship in heterozygote AG genotypes (P.value: 0.064, 95% CI: 0.967-2.981).Conclusion: The frequency of G mutant allele was not statistically significant between the two groups. To ensure the results, it is better to choose a larger statistical society.
