Fabrication and characterization of PCL/Keratin nanofiber wound dressing containing PRF: in vitro and in vivo study

Electrospun nanofibers represent a novel class of materials that show great potential in many biomedical applications including biosensing, regenerative medicine, tissue engineering, drug delivery and wound healing. The aim of this study was to evaluate the wound healing process using polycaprolactone (PCL)/ kreatine (Kr)/ platelet-rich fibrin (PRF) scaffold Prepared by electrospinning method and in vitro and in vivo examinations were done. The morphology of nanofiber dressings was investigated using scanning electron microscopy (SEM) and functional groups and chemical bonds by Fourier transform infrared spectroscopy (FTIR). Then contact angle and degradability were investigated. With the addition of PRF to the PCL / Kr scaffold, the fiber diameter decreased from nm ۱۴۰/ ۰۱± ۳۴ nm to ۶۵/۹۸±۱۴. Biocompatibility was assessed using MTT assay with L۹۹۹ cells and the results showed that PRF-containing dressings improved cell viability. The results of this study show that the scaffold prepared from PCL / Kr / PRF is a suitable candidate for use as a wound dressing as well as for use in skin tissue engineering applications.
