Application of magnetic chitosan nanomaterialsfunctionalized with cysteine, folic acid, and RhodamineB for targeted bioimaging of MDA-MB-۲۳۱ breastcancer cells

Background: Breast cancer is a significant health concernworldwide, and in order to accurately predict and treat cancer,early diagnosis is essential. Despite the availability of variousdiagnostic techniques, magnetic nanoparticles(NPs) are currentlybeing used as an effective cancer diagnostic tool. Indeed,the magnetic properties of the nanoparticles allow for easyseparation of cancer cells from other cells and tissues in orderto make them ideal for bioimaging applications. Specific cellimaging techniques based on luminescent nanocrystals are importantand due to its low cost, high sensitivity and spatial resolution,it has recently played a dominant role in the localizationand dynamic monitoring of disease related biomarkers. Accordingly,the purpose of this study is to assess the diagnostic efficacyof magnetic nanoparticles functionalized with cysteineand folic acid by means of bioimaging techniques.Materials and Methods: Synthesized magnetic nanoparticle(Fe۳O۴@KCC-۱-npr-NH۲) and cysteine were combinedwith magnetic chitosan nanoparticles. The Nano formulationswere characterized for their physicochemical properties usingscanning electron microscopy(SEM) and dynamic lightscattering(DLS). Furthermore, the biocompability and the cytotoxicityof NPs were determined via MTT assay and intracellularuptake of the NPs was observed through fluorescentmicroscope.Results: The result showed that the obtained Fe۳O۴@KCC-۱-npr-NH۲/chitosan-FA/G-GQDs nanoparticles were semi-sphericalin shape and also ۴۸ hours of dose-dependent treatmentrevealed no appreciable loss in cell viability. In essence, upto a concentration of ۴۰۰۰ g ml-۱, these nanoparticles exhibitedgood biocompatibility and no adverse effects were seen inMDA-MB۲۳۱ Breast cancer cells. Additionally, the fluorescentmicroscopy images showed intracellular uptake of the NPs.Conclusion: Due to the fact that folic acid and cysteine arelinked to their receptors -which are expressed at higher levelsin cancer cells compared to healthy cells- Fe۳O۴@KCC-۱-npr-NH۲/chitosan-FA/G-GQDs nanoparticles can be considered aseffective imaging agents in Folic acid expressing cells.
