Background: Breast cancer is the most common cancer among women . Currently, surgery, radiotherapy, and chemotherapyare among the common treatments for breast cancer, which cannotcure the patient in advanced stages and are also harmful toother parts of the body. One of the alternative solutions is to useRNA. RNAs can be used as a widely used research tool due totheir high flexibility and ability to interact with a wide rangeof structures, which inspired the construction of microRNAbindingfragments called sponges. In this project, due to theincreased expression of miR-۲۱ and its importance as a strongoncomir in breast cancer, we sought to reduce the presence ofthis molecule in active pathways in the disease.Materials and Materials and Methods: Breast cancer celllines were cultured in culture media and the expression of miR-۲۱ was measured in them. We transfected the constructs into thecells using liposome ۲۰۰۰ reagent. We measured the expressionof downstream genes related to miR-۲۱ after RNA extractionand cDNA synthesis with PCR and Real time PCR, and finallywe performed MTT assay and scratch assay tests to check cellviability and cell migration.Results: In the cells in which the spongy structure was transfected,the expression of miR-۲۱ and its downstream genes decreased.Comparing the linear structure with its mock showedthe greater impact of the linear structure. The comparison ofthe circular structure with its scramble showed the greater impactof the circular structure. Finally, the comparison of linearand circular structure showed the greater effect of the circularstructure.Conclusion: Considering the circular structure and its higherstability than the linear structure, it can be seen that the circularstructure is preserved for a longer period of time in the cellularenvironment and due to its special structure, it resists cell endonucleasesfor a longer period of time.
