Bioinformatic prediction of miRNAs targetingCCND۱; An approach toward breast cancer therapy

Background: Bioinformatics software are tools designed toextract meaningful information from molecular-biological biology databases and perform sequencing. MicroRNAs (miRNAs)are endogenous small RNAs with diverse functions and play arole in many metabolic and signaling pathways. Dysregulationof miRNAs is a common characteristic of many cancers. As anoncogene, CCND۱ plays a key role in promoting breast cancer.Therefore, identification of miRNAs involved in the regulationof CCND۱ would be useful in finding a treatment approach forbreast cancer.Materials and Methods: The miRtargetlink and targetscandatabases were used to determine miRNAs that interact withCCND۱. The interaction of each predicted miRNAs withCCND۱ ۳´UTR was analyzed by RNAhybrid. Finally, DianamiRpathv۳ was used to find the metabolic and signaling pathwaysrelated to these miRNAs.Results: After analyzing among the miRNAs predicted forCCND۱ by RNAhybrid, three miRNAs that showed a strongerrelationship with CCND۱ were selected, which are hsa-miR-۱۰۶a, hsa-miR-۱۹۳a, hsa-miR-۱۵a. According to the data obtainedfrom Diana tools, a strong relationship between hsamir-۲۰a-۵p and hsa-mir-۱۷-۵p was observed in the signalingpathways (e.g. TGFβ) in cancer.Conclusion: Studies have shown that high expression ofCCND۱ is associated with breast tumor progression. As thehigh expression of CCND۱ is associated with a decrease in thelife expectancy of breast cancer patients, expression modulationof miRNAs mentioned above can be a therapeutic approach toprevent the progression of breast cancer.
